Her younger sister, Preetika Rao, is also an actress and has worked in Indian television.
Rao made her film debut with Ab Ke Baras (2002), directed by Raj Kanwar. Although the film had a modest reception, her performance drew attention from critics and earned her a Filmfare Award for Best Female Debut.
In 2003 she starred opposite Shahid Kapoor in Ken Ghosh's Ishq Vishk, a teen romance produced by Tips Films. The film proved a commercial success and established her as a leading actress in Hindi cinema. The following year she appeared in Farah Khan's Main Hoon Na (2004) alongside Shah Rukh Khan, Sushmita Sen and Zayed Khan, and in the comedy Masti (2004), directed by Indra Kumar.
In 2006 she played the female lead in Sooraj Barjatya's Vivah, a Rajshri Productions release that revolved around an arranged-marriage narrative and was a notable commercial success of the year. She reunited with Shahid Kapoor for the film. In 2008 she played the leading role in Shyam Benegal's social satire Welcome to Sajjanpur, opposite Shreyas Talpade, which was praised by critics. Subsequent appearances include Jolly LLB (2013), directed by Subhash Kapoor, in which she played the wife of the protagonist.
Beyond films, Rao has been associated with public-interest campaigns and endorsements. She co-hosts a podcast titled Couple of Things along with her husband, the radio jockey Anmol Sood (RJ Anmol), focusing on relationships and personal experiences.
Amrita Rao married RJ Anmol, a radio personality, in a private ceremony. The couple announced their relationship publicly only several years after their marriage. They have one son.
Rao is regarded as one of the leading actresses of mainstream Hindi cinema during the 2000s, particularly associated with family-oriented and romantic film genres. Her pairing with Shahid Kapoor in Ishq Vishk and Vivah remains widely referenced in discussions of popular Hindi cinema of the period, and her role in Welcome to Sajjanpur is cited as an example of her work in art-house and parallel cinema.
