The institution is named in honour of Karpoori Thakur, the former Chief Minister of Bihar who was popularly addressed as Jannayak ("leader of the people"). Background

Madhepura is a district headquarters in the Kosi region of Bihar. Historically, residents of Madhepura and surrounding districts such as Saharsa, Supaul, Araria and Purnia depended on medical facilities in Patna, Bhagalpur or Darbhanga for advanced treatment. The establishment of a medical college in Madhepura was intended to address this gap by providing both a teaching institution and tertiary-level healthcare in the region.

The college follows the standard model of government medical colleges in Bihar, offering the Bachelor of Medicine and Bachelor of Surgery (MBBS) programme, supported by an attached hospital that serves as a clinical training site as well as a referral centre for the local population.
